ABSTRACT

ObjectiveThe hygroscopic properties of Mume Flos decoction pieces were studied from the perspectives of macroscopic[water activity(Aw)] and microscopic(water molecular mobility), which provided a theoretical basis for the determination of the safe storage moisture content. MethodAdsorption isotherm of Mume Flos decoction pieces was obtained by static weighing method, and seven common hygroscopic models were fitted and estimated. The best model was selected according to the principle that determination coefficient(R2) was closer to 1, residual sum of squares(RSS) was closer to 0 and Akaike information criterion(AIC) was smaller. According to the optimal model, the absolute and relative safe moisture contents of Mume Flos decoction pieces at 25, 35, 45 ℃ was calculated. Low-field nuclear magnetic resonance(LF-NMR) was used to measure the water molecular mobility in the hygroscopic process of Mume Flos decoction pieces. ResultThe best model to describe the adsorption isotherm of Mume Flos decoction pieces was the Peleg model. According to the model expression, the absolute safe moisture contents of Mume Flos decoction pieces at 25, 35, 45 ℃ were 9.59%, 7.96% and 7.68%, and the relative safe moisture contents were 13.05%, 11.99%, 11.77%, respectively. Mume Flos decoction pieces all contained two water states during the process of hygroscopic absorption at different temperatures, namely bound water T21 and free water T22. During the process of hygroscopic absorption, bound water had the largest increase in peak area. The sum of peak areas of the bound water and free water had a good linear relationship with the moisture contents, and the R2 were 0.959 9, 0.911 8 and 0.974 7 at 25, 35, 45 ℃, respectively. When Aw<0.57, T21 did not change, and the water molecular mobility remained unchanged. When Aw>0.57, T21 showed an increasing trend, and the water molecular mobility increased. The moisture contents of Mume Flos decoction pieces were 8.44%, 6.81% and 6.25% when the water molecular mobility increased at 25, 35, 45 ℃, respectively. ConclusionCombined with the theory of water activity and water molecular mobility, 6.25% is recommended as the safe storage moisture content of Mume Flos decoction pieces, this study can provide reference for determining the safe storage moisture content of other decoction pieces.

ABSTRACT

The aim of this study was to evaluate the variations in the stability and nutrient concentration in concentrates for piglet feeding. Five treatments were established: T1 - Control, standard concentrate formulation (SCF); T2 - PXMore5, SCF with more 5% vitamin-mineral premix (VMP); T3 - PXLess5, SCF with less 5% VMP. All these three treatments used 400kg batches in an INTECNIAL mixer; T4 - FeedMixer, SCF using a 4,000kg batch in an IMOTO mixer; T5 - PremixMixer, SCF using a 1,200kg batch in an MUYANG mixer. For each treatment, bags of 20kg were stored in three storage places for four months where room temperature and relative humidity was recorded daily. The concentration of nutrients was evaluated through centesimal and mineral analysis. The water activity of concentrate was affected by temperature and relative air humidity in different storage places. The greatest variation in concentration of crude protein, mineral residue, copper, zinc, and selenium was due to the PremixMixer treatment. Regarding the guaranteed levels, the critical value was verified only for the chrome concentration.(AU)


O objetivo deste estudo foi avaliar as variações na estabilidade e na concentração de nutrientes em concentrados para alimentação de leitões. Foram estabelecidos cinco tratamentos: T1 - controle, concentrado com formulação padrão (CFP); T2 - PXMais5, CFP com 5% a mais de vitaminas e minerais da pré-mistura (PVM); T3 - PXMenos5, CFP com 5% a menos de PVM (todos os três tratamentos utilizaram lotes de 400kg em um misturador INTECNIAL); T4 - FeedMixer, CFP usando um lote de 4.000kg em um misturador IMOTO; T5 - PremixMixer, CFP usando um lote de 1.200kg em um misturador MUYANG. Para cada tratamento, sacos de 20kg foram armazenados em três ambientes distintos por quatro meses, onde a temperatura ambiente e a umidade relativa do ar foram registradas diariamente. A concentração de nutrientes foi avaliada por meio de análises centesimal e mineral. A atividade de água do concentrado foi afetada pela temperatura e a umidade relativa do ar nos diferentes locais de armazenamento. A maior variação na concentração de proteína bruta, resíduo mineral, cobre, zinco e selênio foi devido ao tratamento "MistPremix". Em relação aos níveis de garantia, foi verificado valor crítico apenas para a concentração de cromo.(AU)

ABSTRACT

Objective:To explore the mechanism of rancidity during storage by researching the changes of water content, relative permeability of cell membrane and rancidity levels of Armeniacae Semen Amarum in deterioration process. Method:Armeniacae Semen Amarum samples under different storage conditions were evaluated and classified by sensory assessors, and samples with different levels of rancidity were obtained. Water content was measured by toluene method, and water activity was obtained by water activity meter. Malondialdehyde (MDA) and relative conductivity were measured using thiobarbituric acid colorimetry and conductivity meter, respectively. The content of fatty oil was obtained by Soxhlet extraction. The acid value and peroxide value were measured in accordance with the general rules 0713 and 2303 of the 2020 edition of <italic>Chinese Pharmacopoeia</italic> (part Ⅳ), respectively. Based on the above experimental data, chemometric methods (cluster analysis, principal component analysis) were selected to establish classification and discriminant models of Armeniacae Semen Amarum with different rancidity levels, in order to verify the accuracy of the classification results. Result:According to the results of sensory evaluation, Armeniacae Semen Amarum samples were divided into three classes, including no rancidity, slight rancidity and rancidity. Compared with the no rancid samples, the color of surface and cotyledon were deepened in rancid samples, and the oil was appeared on surface with rancid taste. The values of water content, water activity, MDA content and relative conductivity were all significantly increased in deterioration process (<italic>P</italic><0.01). The content of fatty oil was significantly decreased with the occurrence of rancidness (<italic>P</italic><0.01), while the acid value and peroxide value increased significantly (<italic>P</italic><0.01). The results of cluster analysis and principal component analysis showed that the rancid samples could be distinguished from the no rancid and slightly rancid samples. Conclusion:The storage conditions under high temperature and high humidity can accelerate the rancidness of Armeniacae Semen Amarum, which is accompanied by the increase of internal water content, the increase of cell membrane permeability and the occurrence of fatty acid rancidity. It is suggested that Armeniacae Semen Amarum should be stored in low temperature, dry environment, as well as short storage time.

【Objective】 To understand the frequency and significance of anti-" Mia" (anti-" Mia" mixtures of antibodies) in local population in Zhongshan, and the influence of different experimental conditions on the activity of human anti-" Mia" . 【Methods】 The microplate-based agglutination assay and polybrene method were used to screen anti-" Mia" in 3 587 blood samples from voluntary blood donors and patients using O type red blood cells with positive Mia antigen, then.rechecked by tube method and microcolumn gel card method. 【Results】 The frequency of anti-" Mia" was 1.06% (38/3 587), among which 60.5% (23/38) were IgM and 39.5% (15/38) were mixture of IgM and IgG; 0.61% (13/2 135) in local blood donors and 1.72% (25/1 452) in patients(P<0.01). 65.8% (25/38) of the population carrying anti-" Mia" had a history of immunity. 57.9% (22/38) were identified to be anti-" Mur" and 42.1% (16/38) anti-" Mia" using GP.Vw erythrocyte. The appropriate incubation time for anti-" Mia" test was 10 min. 【Conclusion】 The frequency of anti-" Mia" was relatively high among blood donors and patients in Zhongshan, and most of the anti-" Mia" carriers had a history of immunity. Most anti-Mia antibodies were active in saline, and some of them were mixture of IgM and IgG. It may be helpful to include Mia positive red blood cells in the irregular antibody screening cell panel to improve the safety of blood transfusion.

ABSTRACT

Objective: The moisture absorption of Rhei Radix et Rhizoma (RRR) with different initial moisture content was studied under the conditions of relative humidity of 20%-85% and temperature of 5, 15, 25 and 35 ℃, respectively, so as to provide reference for the control of safe storage moisture and reasonable storage of RRR. Methods: RRR was stored at temperature of 5, 15, 25 and 35 ℃ and humidity of 45%, 60% and 75%, respectively. The samples were taken at different time points to determine its safe water activity with powder color and mildew as indicators. The isotherm adsorption data of RRR at 5, 15, 25 and 35 ℃ and water activity of 0.2-0.8 were obtained by static weighing method and six isotherm adsorption models, GAB, Oswin, Smith, Halsey, Henderso and Peleg, were used for fitting and evaluation. Results: The absolute safe water activity and relative safe water activity of RRR were 0.5 and 0.6, respectively. The adsorption isotherms of RRR at 5, 15, 25 and 35 ℃ were "S" type, which belonged to type II isotherm. Oswin model was the best fitting model and the model expression was: Meq=A[Aw/(1-Aw)B. According to the model, the absolute safe water content of RRR with different initial moisture content at 5 ℃ was 9.00%, 9.59%, 8.00%, 6.71% and relative safe water content was 10.17%, 10.89%, 9.20% and 8.07%, respectively; The absolute safe water content of RRR with different initial moisture content at 15 ℃ was 8.24%, 8.83%, 7.24%, 5.86% and relative safe water content was 9.57%, 10.17%, 8.59% and 7.20%, respectively; The absolute safe water content of that at 25 ℃ was 7.17%, 7.75%, 5.73%, 4.70% and the relative safe water content was 8.72%, 9.26%, 7.26% and 6.25%, respectively; The absolute safe water content of that at 35 ℃ was 8.00%, 8.45%, 6.53%, 5.21% and the relative safe water content was 9.74%, 9.85%, 8.40%, 7.27%, respectively. Conclusion: Oswin model can be used to predict the equilibrium moisture content of RRR in storage, which can provide reference for the control of safe moisture and scientific maintenance of RRR.

ABSTRACT

Ochratoxin A (OTA) is a mycotoxin produced by filamentous fungi with high impact Lactic acid bacteria; in food safety due to its toxicity. In the last decade, the presence of OTA was widely reported in different foods. In this study, the ability of Lactobacillus (L.) plantarum CRL 778 to control growth and OTA production by Aspergillus (A.) niger 13D strain, at different water activity (a w) values (0.955, 0.964, 0.971, 0.982, and 0.995) was determined in vitro. Both parame ters were significantly (p<0.05) reduced by the lactobacilli and the effect depended on a w. Greatest growth rate inhibition (46.9%) was obtained at a w = 0.995, which is the most suitable value for growth and production of antifungal metabolites (lactic acid, acetic acid, phenyllac-tic and hydroxyl-phenyllactic acids) by L. plantarum CRL 778. Besides, morphological changes and inhibition of melanin synthesis were observed in colonies of A. niger 13D in presence of L. plantarum CRL 778 at a w ranged between 0.971 and 0.995. In addition, maximum reduction (90%) of OTA production took place at a w = 0.971, while inhibition of fungi growth was more evident at a w =0.995. These findings suggest that L. plantarum CRL 778 could be used for control of ochratoxigenic fungal growth and OTA contamination in different fermented foods with a w values between 0.971 and 0.995.


Ocratoxina A (OTA) es una micotoxina producida por hongos filamentosos con un alto impacto en la seguridad alimentaria debido a su toxicidad. En la última década se ha reportado ampliamente a nivel mundial, la presencia de OTA en diversos alimentos. En este estudio se evaluó in vitro, la capacidad de Lactobacillus (L.) plantarum CRL 778 de controlar el crecimiento y la producción de OTA por Aspergillus (A.) niger 13D, a diferentes valores de actividad de agua (a w): 0.955, 0.964, 0.971,0.982 y 0.995). La cepa láctica redujo significativamente (p <0.05) ambos parámetros, siendo el efecto dependiente del valor de a w. La mayor inhibición del crecimiento (46.9%) se obtuvo a a w =0.995, valor más adecuado para el crecimiento y producción de metabolitos antifúngicos (ácido láctico, ácido acético, ácidos fenil-láctico e hidroxi-fenil láctico) por la cepa láctica. Además, se observaron cambios morfológicos en las colonias de A. niger 13D, crecidas en presencia de L. plantarum CRL 778 a valores de a w de 0.971 y 0.995. El porcentaje máximo de reducción en la producción de OTA (90%) por la cepa láctica se observó a un valor de a w = 0.971, mientras la inhibición del crecimiento fúngico fue mayor cuando a w = 0.995. Estos hallazgos sugieren que L. plantarum CRL 778 podría emplearse para el control de la contaminación por hongos ocratoxigénicos en alimentos con valores de aw comprendidos entre 0.971-0.995.

ABSTRACT: Horse leukoencephalomalacia (ELEM) is a disease caused by the ingestion of mycotoxins (fumonisins) produced by fungi of the genus Fusarium that infect corn and/or its byproducts. This disease has been described by ingestion of mature corn with humidity above 15% at temperatures below 20°C. The aim of this paper was to report an outbreak of leukoencephalomalacia in horses fed with immature corn. Two horses out of three showed neurological signs approximately seven days after eating immature corn in its reproductive phase (R2, milky grains). Corn was harvested and administered directly to the animals, with no storage. Deaths occurred approximately 24 hours after the onset of clinical signs. Grossly, there were multifocal dark red to brown areas in the white matter of the telencephalon and hyppocampus and thalamus. Histologically, there was edema and hemorrhage in several areas of the telencephalon white matter, which corresponded to dark red to brown areas observed in the macroscopy. There was also foci of malacia with presence of reactive astrocytes with abundant eosinophilic cytoplasm and inflammatory cells. Diffuse capillary wall degeneration and endothelial cell swelling were also observed. Two ppm of fumonisin were detected by immunoaffinity column method (VICAM) in the immature corn sample. The water activity in this cereal, when the grain is still milky, is 0.98 and can predispose it to growth of mycotoxin-producing fungi. In the present case, fumonisin was found in milky grains in the beginning of the reproductive phase (R2), which suggested that even immature corn may be infected by Fusarium spp. and should not be administered to horses.


RESUMO: A leucoencefalomalácia dos equinos (ELEM) é uma doença causada pela ingestão de micotoxinas (fumonisinas) produzidas por fungos do gênero Fusarium que infectam o milho e/ou seus subprodutos. A doença tem sido descrita pela ingestão de milho maduro com umidade acima de 15% em temperatura ambiente abaixo de 20°C. O objetivo deste trabalho foi relatar um surto de leucoencefalomacia em equinos alimentados com milho verde. Dois equinos de três animais apresentaram sinais clínicos neurológicos aproximadamente sete dias após iniciarem a ingestão de milho verde na fase reprodutiva (R2, grãos leitosos) com palha e talos, colhido no máximo 24 horas antes de ser administrado. A morte ocorreu aproximadamente 24 horas após o início dos sinais clínicos. Macroscopicamente havia no sistema nervoso central áreas multifocais acinzentadas e amareladas na substância branca do telencéfalo, no hipocampo e no tálamo. Histologicamente observou-se edema e hemorragia em diversas áreas da substância branca do telencéfalo, que correspondiam às áreas acinzentadas observadas na macroscopia. Havia, também, próximo as áreas hemorrágicas, focos de malacia com presença de astrócitos reativos com abundante citoplasma eosinophilico e algumas células inflamatórias. Degeneração das paredes dos capilares e tumefação das células endoteliais também foram observadas. Na análise da amostra de milho pelo método de colunas de imunoafinidade (VICAM) foram detectados 2ppm de fumonisina. A atividade de água neste cereal, quando o grão ainda está leitoso, é de 0,98, o que predispõe ao crescimento de fungos produtores de micotoxinas. No presente caso fumonisina foi encontrada nos grãos leitosos no início da fase reprodutiva (R2), o que sugere que mesmo o milho ainda imaturo pode estar infectado por Fusarium spp. e não deve, também, ser administrado aos equinos.

ABSTRACT

Cereal grains are the most important food source for humans. As the global population continues to grow exponentially, the need for the enhanced yield and minimal loss of agricultural crops, mainly cereal grains, is increasing. In general, harvested grains are stored for specific time periods to guarantee their continuous supply throughout the year. During storage, economic losses due to reduction in quality and quantity of grains can become very significant. Grain loss is usually the result of its deterioration due to fungal contamination that can occur from preharvest to postharvest stages. The deleterious fungi can be classified based on predominance at different stages of crop growth and harvest that are affected by environmental factors such as water activity (a(w)) and eco-physiological requirements. These fungi include species such as those belonging to the genera Aspergillus and Penicillium that can produce mycotoxins harmful to animals and humans. The grain type and condition, environment, and biological factors can also influence the occurrence and predominance of mycotoxigenic fungi in stored grains. The main environmental factors influencing grain fungi and mycotoxins are temperature and a(w). This review discusses the effects of temperature and a(w) on fungal growth and mycotoxin production in stored grains. The focus is on the occurrence and optimum and minimum growth requirements for grain fungi and mycotoxin production. The environmental influence on aflatoxin production and hypothesized mechanisms of its molecular suppression in response to environmental changes are also discussed. In addition, the use of controlled or modified atmosphere as an environmentally safe alternative to harmful agricultural chemicals is discussed and recommended future research issues are highlighted.

ABSTRACT

Thai traditional fermented shrimp paste (Ka-pi) is widely consumed as condiment and seasoning ingredient in Thailand. Chemical and physical properties of thirteen shrimp paste samples were examined. The water content and water activity (Aw) of the products ranged from 33.95-52.19 % and 0.64-0.72, respectively while salt concentration was 7.00 to 10.85 %. The nitrogen content varied from 2.87 to 6.85 % (w/w). KS1 showed the highest protein content (42.8 %) followed by SP1 and SP3 (42.4 and 41.3 %), respectively. The predominant amino acids were glutamic acid (70.1-593.9 μg/g), lysine (112.7-546.3 μg/g) and leucine (29.5-544.9 μg/g). Total bacterial cell count in the samples ranged from 1.3x103 - 2.9x105 cfu g-1 while lactic acid bacteria (LAB) were not detected. This study will be provided the information for improving the quality of products in manufacture to meet the demand of consumer.

ABSTRACT

Sorghum, which is consumed in Tunisia as human food, suffers from severe colonization by several toxigenic fungi and contamination by mycotoxins. The Tunisian climate is characterized by high temperature and humidity that stimulates mold proliferation and mycotoxin accumulation in foodstuffs. This study investigated the effects of temperature (15, 25 and 37 °C), water activity (a w, between 0.85 and 0.99) and incubation time (7, 14, 21 and 28 d) on fungal growth and aflatoxin B1 (AFB1) production by three Aspergillus flavus isolates (8, 10 and 14) inoculated on sorghum grains. The Baranyi model was applied to identify the limits of growth and mycotoxin production. Maximum diameter growth rates were observed at 0.99 a w at 37 °C for two of the isolates. The minimum a w needed for mycelial growth was 0.91 at 25 and 37 °C. At 15 °C, only isolate 8 grew at 0.99 a w. Aflatoxin B1 accumulation could be avoided by storing sorghum at low water activity levels (≤0.91 a w). Aflatoxin production was not observed at 15 °C. This is the first work on the effects of water activity and temperature on A. flavus growth and AFB1 production by A. flavus isolates on sorghum grains.


El sorgo, que se consume en Túnez como alimento humano, puede sufrir la colonización severa de varios hongos toxicogénicos, con la consiguiente bioacumulación de micotoxinas. Además, el clima de Túnez, caracterizado por las altas temperaturas y humedad, estimula el crecimiento fúngico y la acumulación de micotoxinas en los productos alimenticios. Este estudio investigó los efectos de la temperatura (15, 25 y 37 °C), la actividad de agua (a w) (entre 0,85 y 0,99) y el tiempo de incubación (7, 14, 21 y 28 días) sobre el crecimiento y la producción de aflatoxina B1 (AFB1) de 3 aislados de Aspergillus flavus (designados como 8, 10 y 14) que se inocularon sobre granos de sorgo. El modelo Baranyi se aplicó para identificar los límites del crecimiento y la producción de micotoxinas. Las tasas máximas de crecimiento para 2 de los aislados se observaron en la combinación 0,99 a w y 37 °C. La a w mínima necesaria para el crecimiento del micelio fue de 0,91 a 25 °C y 37 °C. A 15 °C, solo el aislado 8 creció a 0,99 a w, pero fue incapaz de producir la aflatoxina B1. Es posible evitar la acumulación de aflatoxina B1 en el sorgo almacenándolo a baja actividad de agua (≤ 0,91 a w). Este es el primer trabajo que ha estudiado el efecto de la actividad del agua y la temperatura sobre el crecimiento de aislados de A. flavus y su producción de aflatoxina B1 en granos de sorgo.

OBJECTIVE: To investigate the influence of moisture including water content and water activity on the stability of amoxicillin and potassium clavulanate granules according to the characteristics of the granules, and reveal the key quality control property in the preparation process with stability as an indicator. METHODS: Under the condition of (25±2)℃ and (75±5)% RH, the water content, active ingredient contents, related substances and characteristics of crystal form of amoxicillin and potassium clavulanate granules were studied. RESULTS: The stability of amoxicillin and clavulante potassium granules was positively correlated with water activity in the samples, but was not related to water content closely. The sample with lower water activity was of higher stability. CONCLUSION: The key quality control property of amoxicillin and potassium clavulanate granules is the moisture, and the key process parameter in the preparation is the water activity.

ABSTRACT

Cassava flour from dry and water groups are important starch products widely consumed in the Amazonian region of Brazil. In this study, the moisture sorption isotherms of cassava flour from dry and water groups were obtained in the water activity (aw) range from 0.09 to 0.94 (dry group) and 0.07 to 0.93 (water group) at 25°C and the applicability of eight mathematical models in data prediction was evaluated. Both cassava flour from dry and water groups exhibited type II isotherms (sigmoid) and the hysteresis effect between adsorption and desorption isotherms was shown almost over the entire range of aw. The moisture of cassava flour from dry and water groups should not exceed 11.3g H2O 100g-1 (aw=0.6) to ensure the theoretical microbiological stability and the monolayer moisture value (9.0 and 7.9 g H2O 100g-1, respectively) indicated the level of moisture content to be reached during the drying process to avoid unnecessary power consumption. Finally, the data fit showed that the mathematical model of GAB were able to predict efficiently the sorption isotherms of both cassava flour from dry and water group at the entire aw range.


As farinhas de mandioca do grupo seca e do grupo d'água são produtos amiláceos amplamente consumidos na região amazônica do Brasil. Neste estudo, as isotermas de adsorção e dessorção de umidade foram obtidas para ambos os produtos a 25°C, na faixa de atividade de água (aw) de 0,09 a 0,94 (tipo seca) e 0,07 a 0,93 (tipo d'água). Foi avaliada a aplicabilidade de oito modelos matemáticos na predição das isotermas de sorção. Tanto a farinha tipo seca quanto a farinha tipo d'água apresentaram isotermas tipo II (sigmoide) e um efeito de histerese entre as isotermas de adsorção e dessorção, que se estendeu em toda a faixa de aw. Para que a estabilidade microbiológica teórica seja assegurada, a umidade em ambas as farinhas de mandioca não pode alcançar valores superiores a 11,3 g H2O 100g-1 (aw=0,6). O valor da monocamada das farinhas seca e d'água (9,0 e 7,9 g H2O 100g-1, respectivamente) indicou que esses níveis de umidade não devem ser atingidos durante a secagem dos produtos, para evitar gasto desnecessário de energia. Finalmente, o modelo de GAB mostrou-se altamente eficiente na predição das isotermas de sorção de ambos os produtos, em toda a faixa de aw estudada.

ABSTRACT

Microbial contamination is one of the major inevitable concerns associated with herbal liquid formulations, which may originate from herbal raw materials. Inclusion of preservatives in herbal liquid formulations has been of considerable value for many years. Anti-microbial preservatives are normally added to prevent microbial proliferation while the product on shelf and during in use conditions. The properties of these preservatives are due to certain functional groups, which are usually harmful to living cells and might therefore be associated with certain risks when used in humans and they are the leading causes of adverse reactions and have negative and potentially life threatening side effects, because they not only act on microorganism but may also interfere with human cells. In this study, we have made an effort to develop a preservative-free and self-preserving liquid oral formulation by understanding and applying alternative principles of preservation (approaches other than using preservatives) by taking Ashoka herb extract as a prototype. Our series of formulation trials using different vehicle systems, which reduce the water activity by controlling the pH and osmotic conditions successfully yielded a vehicle system that could be used for the manufacturing of stable preservative-free/self-preserving herbal liquid oral formulations. Ashoka formulations were found to be physically, chemically and microbiologically stable during the six months of accelerated stability studies.

ABSTRACT

O objetivo do trabalho foi avaliar a estabilidade física, físico-química e microbiológica da banana passa orgânica das variedades Caipira, Pacovan Ken e Prata Anã, durante 180 dias de armazenamento, bem como ajustar as curvas de secagem. As bananas foram desidratadas em estufa de circulação de ar forçada a 65°C, até umidade final entre 20-25% (b.u.). A variedade Pacovan Ken atingiu a umidade final após 37h de desidratação. O valor da atividade de água, para as três variedades, manteve-se estável aos 180 dias e caracteriza o produto como de umidade intermediária. Não se observou, em nenhum tratamento, variação significativa dos parâmetros físico-químicos pela análise de variância. Houve escurecimento do produto nas três variedades, com perda significativa do brilho (L*) e da intensidade da cor amarela (b*) e aumento da intensidade da cor vermelha (a*) durante o armazenamento. Houve estabilidade microbiológica do produto, com baixa contagem de bolores e leveduras e ausência de Salmonella e coliformes totais a 35°C em todas as variedades, conforme Legislação vigente. A banana passa orgânica apresentou estabilidade comercial nos meses de estudo.


The objective of this study was to evaluate the physical, physico-chemical and microbiological stability of dried organic banana varieties Caipira, Pacovan Ken and Prata Anã, 180 days of storage, as well as adjust the drying curves . The bananas were dried in an oven with forced air circulation at 65 ° C until final humidity of 20-25 % ( wb). The variety Pacovan Ken reached the final moisture after 37h of dehydration . The value of water activity for the three varieties, remained stable at 180 days and features the product as intermediate moisture. Not observed in any treatment, significant variation of physico-chemical analysis of variance parameters. There darkening of the product in all three varieties, with a significant loss of brightness (L*) and the intensity of the yellow color (b*) and increased intensity of red color (a*) during storage. There was microbiological stability of the product, with low count of yeasts and molds and Salmonella and coliforms at 35°C in all varieties, according to current legislation. The dried organic banana trade showed stability in the months of study.

ABSTRACT

Plant tissues are composed of a watery solution of low molecular weight species, mainly sugars, salts and organic acids, and of high molecular weight hydrocolloids, contained in a water insoluble matrix of macromolecules, mostly carbohydrates. All these constituents interact with water, thus reducing its thermodynamic vapour pressure (aw), with small molecules interacting through polar binding, and large biopolymers through surface and capillary effects. Similarly, some constituents will greatly affect kinetic glass transition temperatures (Tg), while others will not. As regards stability, while microbial and chemical changes are mainly related to aw, structure-related changes such as collapse are dependent on the glass transition temperature, Tg. In simple systems such as juices, both thermodynamic and kinetic approaches, employed respectively for high and low moisture systems, have predictive ability, which can be unified in the concept of “critical aw”. However, in complex, multidomain, multiphase systems, such as vegetables and fruits, where insoluble polymeric phases are present, hydrocolloids such as soluble pectins will only slightly affect Tg and aw, but significantly increase the macro viscosity of the soluble fraction, thereby reducing the tendency to collapse. In such cases the use of Tg as a predictive tool must be considered with care. The interrelationships among these aspects are discussed in detail below.

ABSTRACT

El objetivo de este trabajo fue evaluar 14 aislamientos nativos del género Trichoderma (Tn) obtenidos de la rizósfera del cultivo de caña de azúcar de acuerdo con su efecto de inhibición in vitro en cultivos duales contra la cepa comercial de Trichoderma harzianum (T22). Los aislamientos nativos con mayor índice de inhibición fueron los denominados Palma (T12) y Hubero (T11), con un valor de 2, mientras que T01, T04, T05 y T08 presentaron un valor de 1. Además, el mayor porcentaje de inhibición fue causado por el aislamiento T03, con una media de 92.32 ± 2.64%, sin diferencias estadísticas con los nativos T07, con un valor inhibición de 88.99 ± 1.46%, T01 87.93 ± 2.28%, y T12; 87.71 ± 3.16. En este caso, los aislamientos T07 y T12 mostraron capacidad inhibitoria con mayores atributos para su selección como agentes de control biológico. Al evaluar la tolerancia in vitro de los aislamientos nativos de Trichoderma a la actividad del agua (aw), se observó efecto significativo sobre el desarrollo micelial de los aislamientos de Trichoderma nativo evaluados. Los aislamientos T03 (7919.3 ± 932.53) y T12 (6388.2 ± 623.40) presentaron el mejor desarrollo radial. Estos resultados sugieren que los aislamientos T03 y T12 podrían desarrollarse mejor bajo condiciones de estrés hídrico. Por lo anterior, concluimos que los aislamientos T07 y T12, son los mejores candidatos como agentes inhibidores y se sugiere evaluarlos in vitro o invernadero contra el agente causal de la enfermedad del muermo rojo en caña de azúcar (Colletotrichum falcatum).


The objective of this work was to evaluate 14 native e Trichoderma genus isolates (Tn) obtained from the rhyzosphere of sugarcane plantations in accordance with their inhibitory in vitro effect on dual cultures against a commercial strain of Trichoderma harzianum (T22 strain). The native isolates with the highest inhibition levels were the named Palma (T12) and Hubero (T11), with a type value 2, although the T01, T04, T05 and T08 presented activity type 1. The highest percentage of inhibition of the strain T22 was caused by the isolate T03 (average of 92.32 ± 2.64%), with no statistical difference (P >0.05) from the natives isolates T07 (inhibition average of the 88.99 ± 1.46%), T01 (average 87.93 ± 2.28%), and T12 (average 87.71 ± 3.16%). In this case, T07 and T12 isolates showed inhibitory capacity in more attributes for selection as biological control agents. In assessing tolerance in vitro native isolates of Trichoderma to water activity (aw), we observed significant effect on mycelia growth of native Trichoderma isolates evaluated. In addition, isolates T03 (average 7919.3 ± 932.53) and T12 (average 6388.2 ± 623.40) had the best radial development. These results suggest that the isolates T03 and T12 could develop better under water stress conditions. Therefore, we conclude that the isolates T07 and T12 are the best candidates as inhibitory agents and we suggest evaluating under in vitro condition or in greenhouse against with the causal agent of the red rot disease from the sugarcane (Colletotrichum falcatum).

ABSTRACT

Vibrio harveyi is considered as a causative agent of the systemic disease, vibriosis, which occurs in many biological fields. The effects of temperatures (12.9-27.1 ºC) and water activity (NaCl% 0.6%-3.4%) on V. harveyi were investigated. The behavior and growth characteristics of V. harveyi was studied and modeled. Growth curves were fitted by using Gompertz and Baranyi models, and the Baranyi model showed a better fittness. Then, the maximum growth rates (µmax) and lag phase durations (LPD, λ) obtained from both Gompertz and Baranyi model were modeled as a combination function of temperature and water activity using the response surface and Arrhenius-Davey models for secondary model. The value of r², MSE, bias and accuracy factor suggest Baranyi model has better fitness than Gompertz model. Furthermore, validation of the developed models with independent data from ComBase also shown better interrelationship between observed and predicted growth parameter when using Baranyi model.

ABSTRACT

The surface coverage of certain dry fermented sausages such as Italian salami by some species of Penicillium provides their characteristic flavor and other beneficial properties. One of them is the protective effect by means of a uniform film of white mold against undesirable microorganisms. The aim of this work was to identify and to isolate the fungal species present in mature Italian type of salami and to evaluate if it is possible to obtain some of them as starters. In addition, the effects of temperature (14 °C and 25 °C), water activity (a w) (0.90, 0.95 and 0.995) and 2.5 % sodium chloride (NaCl) on fungal growth were determined. Similarly, the proteolytic and lipolytic activity and the ability to produce toxic secondary metabolites were evaluated in order to characterize some possible starter strain. All species found belong to the genus Penicillium, including a performing starter as Penicillium nalgiovense and some potentially toxicogenic species. All the strains showed a higher growth rate at 25 °C. The production of extracellular proteases and lipases was significantly higher at 25 °C than at 14 °C with and without sodium chloride. Only Penicillium expansum produced patulin. On the other hand, Penicillium griseofulvum was the only species that produced ciclopiazonic acid but none of the strains produced penicillin. The species present on salami, Penicillium nalgiovense, Penicillium minioluteum, Penicillium brevicompactum and Penicillium puberulum were unable to produce any of the evaluated toxins. These findings suggest that some fungal isolates from the surface of salami such as P. nalgiovense are potentially useful as starters in sausage manufacture.


La cobertura de la superficie de los embutidos fermentados secos -como el salamín tipo italiano- por algunas especies de Penicillium les proporciona un sabor característico y otras propiedades beneficiosas. Una de ellas es el efecto de protección contra microorganismos indeseables, al formarse una película blanca uniforme de mohos. El objetivo de este trabajo fue aislar e identificar los hongos filamentosos encontrados en la superficie de salamines tipo italiano y evaluar la posibilidad de obtener especies para utilizarse como cultivos iniciadores. Se determinó el efecto de la temperatura, la actividad de agua y del cloruro de sodio sobre el crecimiento fúngico. La actividad proteolítica y lipolítica y la capacidad de producir metabolitos secundarios tóxicos fueron evaluadas con el fin de caracterizar algunos posibles cultivos iniciadores. Todas las cepas fúngicas aisladas e identificadas correspondieron a especies del género Penicillium, algunas benéficas, como Penicillium nalgiovense, y otras potencialmente toxicogénicas. Estas cepas tuvieron diferentes tasas de crecimiento en respuesta a las diferentes condiciones de cultivo. Todas las cepas mostraron mayor crecimiento a 25 °C. La producción de proteasas y lipasas extracelulares fue significativamente mayor a 25 °C que a 14 °C. Penicillium expansum fue la única especie que produjo patulina y Penicillium griseofulvum fue la única que produjo ácido ciclopiazónico. Ninguna de las especies produjo penicilina. Penicillium nalgiovense, Penicillium minioluteum, Penicillium brevicompactum y Penicillium puberulum no produjeron ninguna de las toxinas evaluadas. Estos resultados sugieren que algunos aislamientos fúngicos, como P. nalgiovense, son potencialmente útiles como cultivos iniciadores en la fabricación de estos productos.

ABSTRACT

The study of thermal resistant mould, including Byssochlamys nivea, is of extreme importance since it has been associated with fruit and fruit products. The aim of this work is to analyze the influence of water activity (a w) and ascospore age (I) on the growth of Byssochlamys nivea in pineapple juice. Mold growth was carried out under different conditions of water activity (a w) (0.99, 0.96, 0.95, 0.93, 0.90) and ascospore age (I) (30, 51, 60, 69, 90 days). Growth parameters as length of adaptation phase (λ), maximum specific growth rate (µmax) and maximum diameter reached by the colony (λ) were obtained through the fit of the Modified Gompertz model to experimental data (measuring radial colony diameter). Statistica 6.0 was used for statistical analyses (significance level α = 0.05). The results obtained clearly showed that water activity is statistically significant and that it influences all growth parameters, while ascospore age does not have any statistically significant influence on growth parameters. Also, these data showed that by increasing a w from 0.90 to 0.99, the λ value substantially decreased, while µmax and λ values rose. The data contributed for the understanding of the behavior of B. nivea in pineapple juice. Therefore, it provided mathematical models that can well predict growth parameters, also helping on microbiological control and products' shelf life determination.

ABSTRACT

The aim of this research was the analysis of the possible antagonistic effect of Penicillium oxalicum over the pathogen rice fungus A. alternata under different conditions of temperature, water activity and culture media. The macroscopic study of the dual growth revealed that according to the Index of Dominance P. oxalicum was more competitive that A. alternata at 25ºC whereas at 15ºC was this species. Microscopic analysis showed that P. oxalicum was a mycoparasite of A. alternata at all conditions tested. The antagonist penetrated into A. alternata and disintegrated its conidiophores and conidia. The results suggests that P. oxalicum may be a possible biological control agent of the rice pathogens in a future.
